對於貼壁(bi)細胞(bao)，傳(chuan)代(dai)可參(can)考以(yi)下方法：

1. 棄去培養上清，用(yong)不含鈣(gai)、鎂離子的(de)PBS潤(run)洗細胞(bao)1-2次。

2. 加2ml消(xiao)化(hua)液(ye)(0.25%Trypsin-0.53mM EDTA)於培養瓶中(zhong)，置於37℃培養箱中(zhong)消(xiao)化(hua)1-2分(fen)鐘(zhong)，然後(hou)在(zai)顯微鏡下(xia)觀察(cha)Ⅱ型(xing)肺(fei)泡上皮(pi)細(xi)胞(bao)|細胞(bao)消化(hua)情況(kuang)，若(ruo)細胞(bao)大(da)部(bu)分(fen)變(bian)圓並(bing)脫(tuo)落(luo)，迅速(su)拿回(hui)操作臺(tai)，輕(qing)敲幾下(xia)培養瓶後(hou)加(jia)少量(liang)培養基終止(zhi)消(xiao)化(hua)。

3. 按(an)6-8ml/瓶(ping)補(bu)加培養基，輕輕打勻(yun)後(hou)吸(xi)出(chu)，在1000RPM條(tiao)件(jian)下離心(xin)4分鐘(zhong)，棄(qi)去上清液(ye)，補加(jia)1-2mL培養液後(hou)吹勻(yun)。

4. 將(jiang)細胞(bao)懸液(ye)按(an)1：2到(dao)1：5的(de)比例(li)分到(dao)新的(de)含(han)8ml培養基的(de)新(xin)皿中(zhong)或(huo)者(zhe)瓶(ping)中(zhong)。

對(dui)於懸浮細(xi)胞(bao)，傳(chuan)代(dai)可參(can)考以(yi)下方法：

方法壹：收(shou)集(ji)細(xi)胞(bao)，1000RPM條件(jian)下離心(xin)4分鐘(zhong)，棄(qi)去上清液(ye)，補加(jia)1-2mL培養液後(hou)吹勻(yun)，將(jiang)細胞(bao)懸液(ye)按(an)1：2到(dao)1：5的(de)比例(li)分到(dao)新的(de)含(han)8ml培養基的(de)新(xin)皿中(zhong)或(huo)者(zhe)瓶(ping)中(zhong)。

方(fang)法(fa)二(er)：可選(xuan)擇半數換液(ye)方(fang)式(shi)，棄去半數培養基後(hou)，將(jiang)剩余(yu)細(xi)胞(bao)懸起(qi)，將(jiang)細胞(bao)懸液(ye)按(an)1：2到(dao)1：3的(de)比例(li)分到(dao)新的(de)含(han)8ml培養基的(de)新(xin)皿中(zhong)或(huo)者(zhe)瓶(ping)中(zhong)。

待細胞(bao)生長狀(zhuang)態良好時(shi)，可進(jin)行293/HEK-293細胞(bao)|細胞(bao)凍(dong)存(cun)。貼壁(bi)細胞(bao)凍(dong)存(cun)時，棄去培養基後(hou)加(jia)入(ru)少量(liang)胰(yi)酶(mei)，細(xi)胞(bao)變圓脫落後(hou)，加(jia)入(ru)約1ml含血清的(de)培養基後(hou)加(jia)入(ru)凍(dong)存(cun)管中(zhong)，再(zai)添加(jia)10%DMSO後(hou)進(jin)行凍(dong)存(cun)。懸浮細(xi)胞(bao)凍(dong)存(cun)時，應將(jiang)細胞(bao)收集，1000RPM條(tiao)件(jian)下離心(xin)4分鐘(zhong)，少量(liang)保(bao)存(cun)上清液(ye)(防止(zhi)細(xi)胞(bao)吸走(zou))，加(jia)入部(bu)分新(xin)鮮培養基，加入到(dao)凍(dong)存(cun)管中(zhong)，在(zai)凍(dong)存(cun)管中(zhong)加(jia)入(ru)10%DMSO後(hou)進(jin)行凍(dong)存(cun)。
